A giant alien spider escapes from a military lab and rampage the city of Los Angeles. When a massive military strike fails, it is up to a team of scientists and one clever exterminator to kill the creature before the city is destroyed.

Alex Mathis (Greg Grunberg - Hollow Man [2000]) is a cash exterminator but he works in Los Angeles. While helping an older woman with a rodent problem, she is bitten by a poisonous spider and treated in a hospital. Meanwhile, a mortified man is bitten by a rodent-sized spider crawling out of a corpse that has just reached the hospital. Learning from this, Alex offers to take care of the spider in exchange for cleaning his hospital bill. José Ramos (Lombardo Boyar - Candyman: Day of the Dead [1999]), a hospital security guard, volunteers to help Alex. On the way to the morgue, Alex deduces that the spider escaped in a vent. He enters the vent and communicates with Joseph via walkie-talkie. Meanwhile, a military task force led by Major Braxton Tanner (Ray Wise - Swamp Thing [1982]) and Lieutenant Karly Brant (Clare Kramer - The Lost Tree [2015]) are presented at the hospital to examine the dead body of the spider exploded.

Meanwhile, the spider, which grows exponentially, kills a crippled hospital patient. Again inside the ventilation, Alex stumbles upon a cobweb and learns that it is highly flammable. Alex ventures into the basement of the hospital, where he has a confrontation with the spider. The spider is expelled by Karly and escapes into a grate down the sewer, where she kills a homeless man. Alex, in love with Karly, offers his help to Braxton, who declines, especially when Alex asks him for answers about the origin of the spider. Jose convinces Alex to continue his quest, as it could be the most important thing in their lives.

The duo follows the spider still growing in a public park, where it kills dozens of people. Alex with Joseph get the spider to chase their truck and lure the military. A huge shootout occurs, but the spider escapes in the nearby woods. Karly and a group of soldiers chase after him, but the spider ambushes them, kills the majority of the soldiers and captures Karly. After Alex and Joseph asked for answers, Braxton revealed that they discovered the DNA and hoped to use it as a growth hormone. However, a spider's nest was hidden in some test fruit, thus exposing the spider to DNA and activating its rapid growth. It is also revealed that the spider will soon begin to reproduce based on its size and its studies of alien DNA.

Alex and Joseph are heading towards the city, while the gigantic spider installs a nest in the tower of the Bank of the United States. Upon entering the skyscraper, Braxton informs them that Karly is directly inside the nest, and warns them of an imminent aerial attack. They arrive at Karly and free her and several other trapped citizens just before the birth spiders can attack. The group does it outside the building seconds before the air strike strikes, provoking the spider. Alex deduces from the flammability of the cobwebs that the spider could be killed by firing its row. Joseph takes a rocket launcher from a military vehicle and throws it at Alex, who shoots the spider in the row. The spider explodes, finally killing her. Alex hugs Karly, and they both enjoy a passionate kiss.

In a mid-credit scene, Braxton calls Alex to tell him they have another problem. In the background, a giant cockroach can be seen climbing the Statue of Liberty on a video monitor.

Big Ass Spider! is a 2013 science fiction comedy-horror film directed by Mike Mendez. The film tells the story of a bug exterminator, who, with the help of a hospital security guard and the military, battles a giant spider that goes on a rampage in Los Angeles.
